3D Cell Culture and Analyses of Cancer Cells

Abstract

3D cell culture provides a closer model to tumors than the conventional 2D cell culture. In this project, 3D cell culture was done for the hepatocellular carcinoma liver cancer cell lines HepG2 and HUH7. The cell lines compatibility with 3D applications were analyzed visually through light microscopy and fluorescence. The spheroid size is easily adjusted with different cell densities and droplet volumes, as outlined in the protocol. Spheroids were analyzed through visual inspection, light microscopy, fluorescence imaging, and flow cytometry. The viability change of cells after being cultured into spheroids was analyzed with a cell counting machine. Details of 3D cell culture and microscopic analyses of cancer cell spheroids will be discussed.
